En la mitología griega, Mitilene es una hija del rey Macareo de Lesbos, un helíada que colonizó la isla cuando huyó de su patria, Rodas, por haber dado muerte a su hermano Ténages. Algunos autores la hacen hija de Pélope. Fue amante del dios Poseidón, que la hizo madre de Mitone. Una tradición la consideraba fundadora epónima de la ciudad de Mitilene, aunque algunos afirmaban que el nombre deriva del de su hijo o de otro personaje llamado Mitile.
